After measuring caffeine levels in the subjects' saliva, the researchers asked everyone to look at some photos. Five minutes later, some people were given pills containing 200 milligrams of caffeine (equivalent to the amount of caffeine in a large cup of coffee), while others were given a placebo pill without caffeine. Saliva was tested again 1 hour, 3 hours and 24 hours after taking the pill.

The study aimed to assess the relationship between coffee and caffeine use and suicide rates in three large cohorts in the United States. Researchers from Harvard University obtained data from 43599 men from the Health Professionals follow-up study (HPFS,1988-2008), 73820 women from the Harvard Nurses' Health Research Program (NHS,1992-2008) and 91005 women from the Harvard Nurses' Health Research Program Ⅱ (NHS II, 1993). Subjects' use of caffeine, coffee and non-caffeinated coffee was assessed using a certified dietary frequency questionnaire every four years, while suicides were determined by clinicians based on death data. The researchers used the Cox proportional hazard model to estimate the multivariate adjusted relative risk (RRs) and the random effects model to summarize the RR values of each cohort.
